Evaluation of gene expression and test of new therapies by preclinical imaging in murine models

In the last years murine models have become interesting tools to study gene expression and to test innovative classes of targeted therapeutics. RNA-mediated gene silencing and gene therapy hold particular interest in oncology, cardiovascular research, and in the bone regeneration field. Small-animal imaging has been an area of intense worldwide development in the last years, allowing to monitor changes at the cellular and molecular level in living animals, and refine preclinical research procedures, in agreement with the 3Rs principles. We demonstrated that morphological and functional imaging techniques are complementary tools to conventional in vitro methods for reflect the inhibition of tumor growth by RNA interference therapies, or quantify ectopic bone formation induced by virus-mediated gene delivering in living animals. We showed that in these circumstances, imaging allows to perform longitudinal studies, improving the accuracy of experimental results and reduce the number of animals involved into the research.

PET-CT fusion images with 3D volume rendering and simultaneous display of areas of FLT uptake in a medulloblastoma murine model: a wider bone defect in the occipito-parietal portion of the skull is visibile in the ADV5 MOCK medulloblastoma, with higher [18F]-FLT uptake; in AdV5-199b transfected tumor, the size was smaller and radiotracer uptake was lower.
