Description:
Active targeting based on the ligand-target affinity is a common strategy to enhance the binding and the uptake of theranostic nanosystems  into tumoral tissues and microenviroment.
Peptides are widely employed as targeting ligands for their several advantages including the capability to selectively recognize specific diseases markers, and to be easily manipulated for achieving the decoration of smart nanosystem surface. Carbohydrates can also potentially be considered good candidates although the low resistance to glycosidases and the low protein affinity narrow their use as native compounds. Recently we focused on developing novel peptides and carbohydrates for the decoration of nanoparticles (i.e TiO2/Fe2O3, SPION/Au heterostructures), prepared at IBCF-CNR and University of Bari, with integrated multifunctionality for theranostic applications. Both chimeric peptides, displaying motifs for efficient integrins targeting and surface capping function, and thio-glycosides, featuring the sulfur atom at anomeric position for improving glycosidase stability and asialoglycoprotein receptor specific targeting, were designed. These molecules are chemically synthesized and characterized by means of spectroscopic and biological techniques.
